extract biological material that hasnt been studied yet and test it by ingestion.
most common plants have been done, but you could find something novel with enough persistence. fungi might work, as could animals that are poisonous or venomous in some form.
the biggest open field is probably marine reef organisms. they have a lot of biochemistry going on that isnt well studied.
when you found something psychoactive, put the extract into a gas chromatograph and try to figure out what the active compound might be.
any of this can easily kill you. be sure to leave coherent notes so someone can figure out wtf happened to you. good luck

you read the literature and adapt it to the equipment you have in your lab, keeping in mind the stuff they do in academic papers is overkill if you just want product like schlenk techniques, high vaccum, or chromatography columns.
