Is it possible that the Borg were initially notified about Earth and humans after their encounter with the crew of the NX-01 Enterprise (and transmitted the encounter to Unimatrix-1 in the Delta Quadrant), and the Borg Cube that appeared at Wolf 359 was the SAME Cube that Enterprise-D 'encountered' when Q sent them into direct contact with them, albeit pursuing at sub-warp speeds?

Its possible but not likely. The borg canon is a bit of a mess. But its likely the Borg have known about humanity for a long time but didn't care about them. They only made an effort to invade when the Enterprise first encountered the one Cube.
From the Borgs pov, a random star ship on the wrong side of the galaxy, lacking the technology to get there and put up a fight against the Cube, suddenly vanished. Of course thats going to make the borg scratch their heads wondering what the hell is going on.
I don't think the borg have any real interest in humanity, instead seeing them as a stepping stone to understand the Q.

The cube in best of both worlds, data says it matches the exact specifications from the cube during Q's intervention.
Crazy to think it's just been traveling 7 years non-stop, doing absolutely nothing like a baby.
